Secondary Metabolite Production in Callus Culture of <i>Verbascum scamandri</i> Murb.

<i>Verbascum scamandri</i> Murb. known as "Kazdağı Mullein" is an endemic species in Türkiye and classified endangered (EN) species. The aim of this study to establish efficient callus culture for <i>V. analyze the amounts verbascoside, luteolin, aucubin metabolites calli samples. Leaf explants were cultured on MS medium with cytokinin (BAP, Kin, 0, 0.5, 1, 2, 3 mg/L) auxin (NAA, 2,4-D, 0.1, 1 mg/L), g/L PVP, 3% sucrose, 0.7% agar induction. Callus tissue 2 mg/L 0.5 Kin+0.5 2,4-D was proliferated basal containing PGR at same concentrations combinations induction media. Verbascoside, quantified leaf samples the<i> vivo</i> collected plants, <i>in vitro</i> growing using HPLC-DAD. According results, verbascoside content plants 7.03 mg/g, luteolin 0.66 2.99 mg/g. had 1.62 mg/g 0.18 1.32 aucubin. Whereas, maximum secondary observed 13.77 0.51 9.32 2,4-D.
